Immunity, hematopoiesis and genome evolution in zebrafish.
identified Stat1b as a new anemia related factor, showing that abnormality in immune system can lead to aberrant red and myeloid blood cells differentiation. Predicted stat1b in zebrafish by BLASTing the zebrafish stat1a sequence in NCBI genomic database. Cloned the full-length cDNA of stat1b by RT-PCR. Performed exon/intron analysis for stat1b in Ensembl. Made genomic synteny comparison to investigate the evolution history of STAT superfamily. Showed that loss of stat1b function increased the number of red blood cells, while depressed myeloid cells by RNA in situ hybridization and quantitative PCR.

Image analysis for hemoglobin staining

Established a fast and objective method to identify anemia phenotypes in computer programs. Prepared o-dianisidine staining for hemoglobin in wild type and FA mutant zebrafish. Wrote programs in Python and Matlab for image analysis. Applied green color channel to isolate a fish sample from experience and red color channel for hemoglobin staining quantification. Verified a lower hemoglobin staining, ie, anemia, in FA mutant zebrafish.

Biology assay and statistical interpretation of a clinical small molecule screen in zebrafish.
Designed de novo the screen strategy and wrote a grant proposal for a small molecule screen to identify therapeutic agents for Fanconi Anemia. Established a quantitative acridine orange spectroflurometry as biology assay. Determined the number of zebrafish embryos needed by power analysis. Interpreted the results by statistical analysis.

Contributed to establishing the new theory that small molecular RNAs, neglected, play important roles in regulating development. Discovered changes in expression profiles of small molecular RNAs in differentiated PC12 cell a neuron cell line by high resolution SDS-PAGE.
Proved that the gene of GDNF might alleviate Parkinson Disease. Demonstrated that MN9D cell line transfected with the GDNF was resistant to toxic drugs that induce Parkinson Disease.
